Abstract. The cyclodextrin glycosyltransferase (CGTase) is an important enzyme for cyclodextrin (CD) production, and is also widely used in the biotechnology, food, and pharmaceuticals industries. Secretory CGTase production by recombinant Komagataella phaffii using defined medium is a promising approach because of low cost, less impurity protein. CGTase overexpression enabled a burst of reactive oxygen species production and activated pathogenesis-related gene expression, indicating that the transgenic cotton was better prepared to protect itself from infection.
Production of cyclodextrin glucanotransferase CGTase production. CGTase was purified around 20.21 (CGTase) from alkalophilic Bacillus sp. TS1-1: Media fold with a yield of 55.14%. Molecular weight of the optimization using experimental design. Enz.
A low level (0.75%) of nitrogen source was also reported as the CGTase from B. macerans initially favors α-CD production; only in later stages of the reaction does the yield of β-CD approximate or exceed that of its α-homolog. Keywords Activate Charcoal Conversion Reaction Azeotropic Distillation Debranching Enzyme Guest Compound Cyclodextrin glycosyltransferase (CGTase) catalyzes starch conversion into cyclic or linear oligosaccharides, important industrial products for the complexation of non-polar substances.
Production of a novel cyclodextrin glycosyltransferase (CGTase) from Klebsiella pneumoniaeAS‐22 strain, which converts starch predominantly to α‐CD at high conversion yields, in batch, fed‐batch, and continuous cultures, is presented.
First starch is liquified either by heat treatment or using α-amylase, then CGTase is added for the enzymatic conversion. The industrial production of CGTase was made attractive only when alkaliphilic Bacillus species were introduced as producing organism (23). This paper reports the production optimization and some biochemical properties of a CGTase produced by a strain of Bacillus licheniformis isolated from cassava culture soil. production of CGTase, different parameters such as incubation periods (0-72 h), medium pH (9, 9.5, 10, 10.5, 11 and 11.5) and temperature (28ºC, 32ºC, 37ºC, 42ºC, 47ºC and 52ºC) were used. The influence of various carbon and nitrogen sources for the maximum production of CGTase production was studied. The carbon sources Effect of concentration of C, N and sodium carbonate: The effect of different concentration of sago starch on CGTase production was observed.

Enz. 2009-07-08 · CGTase productionThe experiments were made using a CGTase from B. circulans DF 9R, isolated from rotten potatoes and purified by affinity chromatography on α-CDs coupled to Sepharose-4B . The microorganism was cultured in a minimum saline medium for enzyme production as described in a previous report . 2.3. CGTase cyclizing activity Background. Cyclodextrins (CD) are cyclic oligosaccharides derived from starch.
CGTase production was carried out using mL
Production of CGTase by B. circulans P28 in 2 L stirred tank bioreactor increased propotionally with the increase in agitation speed, ranging from 400 to 900 rpm though growth was slightly inhibited at agitation speed of above 600 rpm. The analysis of production of CDs from 1% soluble starch by the wild-type CGTase and its mutants at pH 7.5 showed that higher γ-CD–forming activity leads to greater γ-CD production. In the case of the A223H, A223K and A223R mutants, the γ-CD production increased but there was little change in the β-CD production.

CGTase overexpression enabled a burst of reactive oxygen species production and activated pathogenesis-related gene expression, indicating that the transgenic cotton was better prepared to protect itself from infection.
The maximum CGTase activity obtained on SC was 1,155 U mL(-1) under aerobic conditions. CGTases are produced by a variety of bacteria, mainly Bacillus species, by submerged culture in complex medium. CGTases differ in the amount and types of CDs produced. In addition, CGTase production is highly dependent on the strain, medium composition and culture conditions.
